I'm a fresh from the factory kind of guy and I am in the textiles field. You should wash towels before you use them because they are still coated in some finishing products that help them appear more luxurious, shiny and softer. However it is usually includes at least some wax which blocks water absorption meaning you'll still be wet when you're done drying off. Also if you wash your towels seperately, don't use a dryer sheet. These things are great but they work by locking in moisture so that static electricity charges can be distributed evenly over the surface of the towel instead of in small areas which cause static cling. However in the case of the towel this moisture lock in is not a desirable attribute. I don't think underwear, especially tidey whities have anything on them you should be concerned about. As long as they are from a reputable company in America.
