SpeedRacer 1 #26 September 5, 2003 Here's a recipe for making a polyacrylamide gel for sequencing DNA: 36 gm Urea 1 gm Resin 10 ml 10X Tris-Borate-EDTA (TBE) buffer 52 ml distilled H20 10 ml 10X Polyacrylamide solution Drop in stir bar and stir until urea is dissolved. Vaccum filter through a 0.2 micron filter & de-gas 5 to 10 minutes. Add 500 microliters 10% Ammonium Persulfate and 70 microliters Temed. Swirl gently to mix. Draw up solution in a 50 ml syringe and inject between sequencing plates, being careful not to let bubbles form. Clamp loading comb in place. Let polymerize 2 hours. Place in gel rig and fill reservoirs with TBE buffer (1X). Dissolve sequencing reactions in 20% Gel Loading Buffer, 80% Formamide. Heat to 95 degress C 5 minutes & place on ice. lawrocket 3 #28 September 5, 2003 Honkey Carne Asada This is a little recipe I cam eup with when, in a fit of curiosity, I decided to try somethign different. Take some round steaks and set aside. Slice a white onion and a Bell pepper. Chop quite a bit of cilantro. Set aside. Mash up a papaya (nature's meat tenderizer). Pour the papaya into about a pint of orange juice or Sunny Delight. Add some crushed pineapple to it. Pour everything you've got into the ziplock. Add some hot peppers of your choice and about a tablespoon of cinnamon. Finally, add a few crushed cloves of garlic. Shake it up good. Add a little soy sauce and then drop in the meat to marinate for a few hours. Barbecue the meat. It sounds gross, but it tastes mighty nice!!! My wife is hotter than your wife. (about 36) jumbo chicken wings 3 tbsp. lemon pepper 3 tbsp. vegetable oil 1 cup honey 3/4 cup prepared mustard 1/2 cup lime juice 1/4 cup gold tequila 3 tbsp. chopped fresh cilantro 2 tbsp. hot sauce 1. Preheat grill to medium. Trim wings tips from wings, cut through the joint to separate winglets from drummette. Place wing pieces in large bowl, toss with lemon pepper and vegetable oil. 2. Place seasoned wings in a grill basket**; grill for 10-12 minutes per side, turning every 5-6 minutes, or until fully cooked, golden brown, and crisp. 3. To make margarita sauce, in large bowl, whisk together honey, mustard, lime juice, tequila, cilantro, and hot sauce; set aside. 4. Carefully remove wings from grill basket and add to margarita sauce.
